Job Description The Applied Genomics, Computation & Translational Core is looking for a Biomedical Technician to join the team! The Cedars-Sinai Applied Genomics, Computation, and Translational Core (AGCT Core) is a fully equipped, state-of-the-art genomics facility offering data generation and interpretation for basic science and translational research in next-generation sequencing technologies, including single cell omics, spatial transcriptomics, and bulk cell omics. We specialize in single cell omics and spatial transcriptomics via the 10x Genomics, Parse Biosciences, Mission Bio, and NanoString platforms. Our R&D team offers sample preparation for single cell assays, such as nuclei isolation from frozen tissue or single cell isolation from fresh tissue dissociation. The AGCT Core also offers comprehensive services for standard bulk cell sequencing in genomics, transcriptomics, epigenomics, and metagenomics. To learn more please visit?? The Genomics Core Biomedical Technician participates in wet- and dry- laboratory projects and may execute tasks requiring compliance with all Federal and State accrediting agency requirements and/or Good Laboratory Practices (GLP). Responsible for assigned wet lab work, lab maintenance, and writing and recording controlled documents. Performs all activities strictly according to SOPs, with accurate documentation, and may assist quality control team with required activities. Installs and maintains laboratory equipment to ensure proper functioning of devices. Assists in addressing complex issues and troubleshooting. Department-Specific Responsibilities: Perform manual library preparation for bulk cell, single cell, and spatial genomics. Understand and perform all aspects of NGS library sequencing, including library QC analysis, checking index overlap, determining sequencer loading concentration, determining sequencing run parameters, and determining reads needed. Consult with technical support on library preparation or sequencing run performance issues. Understand the chemistry of all NGS assays performed in the Core. Troubleshoot library preparation, sequencing, and equipment issues. Work closely with and guide RA I on performing and analyzing sample and library quantification and QC, and automated library preparation for service requests. Perform sample preparation for single cell assays. Perform and analyze NanoDrop quantification, Qubit quantification, Bioanalyzer QC, and TapeStation QC. Perform Illumina Sequencing by Synthesis sequencing, and understand and analyze sequencing run metrics. Perform DNA/RNA isolation from various sample types. Perform fold dilution, normalization, and conversion calculations. Liaise with researchers on quantification-, QC-, and sequencing-only requests, as needed. Use iLab as a laboratory information system for service requests and billing. Edit charges, as needed, to reflect the work performed as opposed to the work requested. Use LabArchives as an electronic laboratory notebook to record all information and data generated. Use Trello to track projects, record work performed, and plan work schedule. Update pertinent information to LabArchives and/or Trello on a daily basis, Understand how to choose the appropriate reagent kit to use based on sample quality and concentration. Properly handle and work with DNA and RNA samples. Understand the space layout of the lab and what work is performed in what space. Receive and store samples, and document receipt. Perform automated library preparation. Unpack orders and appropriately store reagents according to temperature requirements. Request reagents and consumables for order as needed. Understand next-generation sequencing (NGS). Report results and project status on a daily basis and at weekly lab meetings. Prepare PowerPoint presentations on NGS topics to present to team members. Maintain general lab cleanliness and organization, including thawing freezers and cataloging sample boxes. Attend seminars/webinars as assigned. Attend lab meetings as assigned. Perform other duties as assigned by managers. Qualifications Education: Associate Degree in biology, biochemistry, or related science/engineering field is required. Bachelor’s degree is preferred. Experience & Skills: One (1) year of experience working in a lab or biotech environment. One (1) year of experience working in an aseptic environment, experience working in a biosafety cabinet, and experience working with human and animal tissues. One (1) year of experience maintaining and/or using biomedical equipment for testing. Collaborates to problem solve and make decisions to achieve desired outcomes. Responds timely, effectively and appropriately to deliverables. Establishes effective working relationships with cross-functional team(s).
